A genetic disorder is a health problem caused by one or more abnormalities in the genome. It can be caused by a mutation in a single gene (monogenic) or multiple genes (polygenic) or by a chromosomal abnormality. Although polygenic disorders are the most common, the term is mostly used when discussing disorders with a single genetic cause, either in a gene or chromosome. The mutation responsible can occur spontaneously before embryonic development, or it can be inherited from two parents who are carriers of a faulty gene or from a parent with the disorder. When the genetic disorder is inherited from one or both parents, it is also classified as a hereditary disease. Some disorders are caused by a mutation on the X chromosome and have X-linked inheritance. Very few disorders are inherited on the Y chromosome or mitochondrial DNA.

जीवों में नर के शुक्राणु तथा स्त्री की अंडकोशिका के संयोग से संतान की उत्पत्ति होती है। शुक्राणु तथा अंडकोशिका दोनों में केंद्रकसूत्र रहते हैं। इन केंद्रकसूत्रों में स्थित जीन के स्वभावानुसार संतान के मानसिक तथा शारीरिक गुण और दोष निश्चित होते हैं। जीन में से एक या कुछ के दोषोत्पादक होने के कारण संतान में वे ही दोष उत्पन्न हो जाते हैं। कुछ दोषों में से कोई रोग उत्पन्न नहीं होता, केवल संतान का शारीरिक संगठन ऐसा होता है कि उसमें विशेष प्रकार के रोग शीघ्र उत्पन्न होते हैं। इसलिए यह निश्चित जानना कि रोग का कारण आनुवंशिकता है या प्रतिकूल वातावरण, सर्वदा साध्य नहीं है। आनुवंशिक रोगों की सही गणना में अन्य कठिनाइयाँ भी हैं। उदाहरणार्थ; बहुत से जन्मजात रोग अधिक आयु हो जाने पर ही प्रकट होते हैं। दूसरी ओर, कुछ आनुवंशिक दोषयुक्त बच्चे जन्म लेते ही मर जाते हैं।
